Orchard Therapeutics Publishes Gene Therapy OTL-101 for the Treatment of ADA-SCID Phase 1/2 Clinical Trial Data Data from a Phase 1/2 clinical trial in patients with combined immunodeficiency (ADA-SCID) were published in the New England Journal of Medicine (NEJM). Results of the trial showed that patients had a 100% survival rate for 2-3 years after receiving a single gene therapy treatment, and more than 95% of patients did not need other treatments to control their disease. This long-term efficacy result demonstrates the potential of gene therapy to functionally cure rare genetic diseases, the press release noted.

New coronavirus Accurate and complete gene annotation map completed After the genomics research, they drew the most accurate and complete gene annotation map of the new coronavirus to date, confirmed several protein-coding genes, and found that some genes did not encode any proteins. In addition, they analyzed nearly 2,000 mutations produced by different strains of the new coronavirus, allowing them to better assess the importance of these mutations.

Jiang Rui/ Wang Yongxiong team published a deep learning method for single-cell data analysis in Nature sub-journal

May 10, Jiang Rui team from Tsinghua University Automation Department and Stanford The team of Wing Hung Wong from the Department of Statistics of of the University jointly published an article entitled Simultaneous deep generative modelling and clustering of single-cell genomic data in Nature Machine Intelligence, proposing a new single-cell data analysis method scDEC. The theoretical basis of the

scDEC method is the neural network probability density estimation theory Roundtrip proposed by the team earlier for high-dimensional data (Liu et al, Proc Natl Acad Sci USA, 118(15):e2101344118, 2021). Guided by this theory, scDEC cleverly uses a set of generative adversarial networks to map high-dimensional single-cell data to a low-dimensional latent space, perform cluster analysis in the low-dimensional space, and then use another set of generative adversarial networks. The network maps low-dimensional data back to a high-dimensional space. Through the reciprocating cycle and co-optimization of such two sets of generative adversarial networks, scDEC integrates the identification of cell types and the dimensionality reduction, generation, and batch effect removal of single-cell data, not only realizing the fusion of single-cell gene expression and chromosome opening The identification of cell types based on sex data has also been successfully applied to downstream cell function modeling studies such as inference of cell developmental trajectory and analysis of intracellular gene regulation mechanisms.

West Lake University Tao Liang’s team identified the host of Clostridium knowlesi alpha toxin for the first time Receptor

On May 10, Tao Liang's team from West Lake University published a research paper online entitled "Sulfated glycosaminoglycans and low-density lipoprotein receptor mediate the cellular entry of Clostridium novyi alpha-toxin" in Cell Research. A guided KO screen identified the sulfated glycosaminoglycan as a key attachment factor for Tcnα. Sulfated glycosaminoglycans are ubiquitous on the cell surface and thus serve as ideal landing sites for a variety of infectious agents, including the emerging virus SARS-COV-2 that caused a global pandemic.

This study further revealed that the synergistic effect between sulfated glycosaminoglycans and LDLR family members contributes to the entry of Tcnα. Finally, this study demonstrates that sulfated glycosaminoglycans are key attachment factors for Tcnα-induced myopathy in vivo, and that SCD effectively reduces Tcnα-induced tissue damage in a mouse model. These findings suggest that blocking the interaction between Tcnα and sulfated glycosaminoglycans may be a new strategy for developing potential therapies for Tcnα-induced diseases.

5月11日, Biotech unicorn Ginkgo Bioworks announced an agreement with SPAC company Soaring Eagle Acquisition to go public through a reverse takeover. The rationale behind the

Ginkgo business is that biology can be "programmed" in a computer-like fashion, simply by sequencing the four basic chemical building blocks (ACGTs) of using DNA. The company can engineer mammalian, bacterial and yeast cells for specific purposes. To help alleviate the shortage of diagnostic tests during the pandemic, Ginkgo has also repurposed technology to read and modify living cells.
