Since the outbreak at the end of 2019, the new coronavirus (SARS-CoV-2) has posed an extremely serious threat to human life and health and the stable development of society. Therefore, the search for broad-spectrum and high-efficiency anti-coronavirus drugs has become a key scientific problem in this field that needs to be solved urgently. Recently, the team of Professor Shuwen Liu from the Southern Medical University School of Pharmacy and Fudan University and Guangzhou Medical University have collaborated to elucidate the role and mechanism of acid anhydride-modified proteins against the new coronavirus. The relevant results are in the Nature sub-publication Signal Published online on Transduction and Targeted Therapy (IF: 18.187).

The paper is entitled "Repurposing of a clinically used anti-HPV agent to prevent and treat SARS-CoV-2 infection as an intranasal formulation". Professor Jiang Shibo of Fudan University, Professor Yang Zifeng of Guangzhou Institute of Respiratory Health/State Key Laboratory of Respiratory Diseases, Guangzhou Medical University, and Guangzhou National Laboratory are the co-corresponding authors of this article. Professor Jiang Shibo of Fudan University and Southern Medical University Professor Shuwen Liu in an early collaborative study found that many common animal or human proteins can be modified by acid anhydrides to antagonize the invasion of multiple viruses through the effect of positive and negative charges. Among them, β-lactoglobulin (β-LG) extracted from milk has been modified by 3-hydroxyphthalic anhydride (3HP), and 3HP-β-LG has been successfully developed as an anti-HPV biological protein dressing (National Second Class medical equipment), used in more than 1,100 hospitals in 23 provinces, municipalities and autonomous regions across the country, and has a positive effect on high-risk HPV infections in the reproductive tract.

3HP-β-LG's anti-SARS-CoV-2 effect and mechanism of action B.1.1.7, B.1.351, P1, B1.617.1 and other SARS-CoV-2 mutant pseudoviruses with strong sex and low neutralizing antibody sensitivity also have an inhibitory effect. The mechanism of action of the acid anhydride modified protein, through experimental studies, is speculated to be that 3HP-β-LG binds to the new coronavirus S protein, competitively blocking the binding of the S protein to the ACE2 receptor, thereby inhibiting the virus from invading target cells. Therefore, 3HP-β-LG is very likely to be developed into a new class of cheap and safe anti-new coronary pneumonia drugs delivered through the nose, providing a new and powerful means for clinically fighting the new crown epidemic.
