Pfizer and Moderna mRNA vaccines shine in this new crown epidemic, giving many people an illusion: mRNA vaccine is a good vaccine.
But in fact, Germany once developed an mRNA new crown vaccine, but it was not approved for marketing because of its poor clinical protection effect.

Because the protective effect of vaccine depends on its antigen design.
The side effects of vaccines are determined in many ways.
1, side reaction problems of mRNA vaccine
In a sense, the current mRNA vaccine is not the most ideal vaccine.
The reason is very simple. After young people get mRNA vaccine, they have a high fever rate. This leads to it being a vaccine that young people don’t like.
For example, the vaccination rate of young people in many European and American countries is relatively low. The reason is that "the fever after vaccination is too uncomfortable", so many young people resist vaccination.
Their elderly have a high vaccination rate, and the reason is that the elderly have a low fever rate after receiving mRNA vaccine.
Although the protection efficiency of inactivated vaccine is not good enough, the rate of fever after vaccination is much smaller than that of mRNA vaccine.
However, fever after vaccination is not a very clinically significant side reaction because it does not have substantial damage to the body.
. If you look at the side reactions that are truly meaningful, there is no big difference between inactivated vaccine and mRNA vaccine.
For example, the incidence of pericarditis and myocarditis caused by mRNA vaccine is actually very low (≤40 of 100,000). This side reaction mainly occurs in people aged less than 40, and is basically a transient reaction and usually has no serious consequences.
Statistics from Hong Kong confirm that there is no difference between the Pfizer vaccine and the inactivated vaccine for relatively serious and meaningful side reactions.
Looking back, the fever caused by mRNA vaccines will definitely become an important basis for "vaccine hesitation", "vaccine suspicion" and "vaccine rumors".
Although a data-looking party like me doesn't think it's such an important thing. But it will definitely make ordinary people feel fear, worry and suspicious.

2. Why is Pfizer and Moderna's mRNA better protective effect?
is different from what many people imagine. In fact, the current mainstream Pfizer vaccine, the excellent protective effect of the Moderna vaccine does not come from mRNA.
mRNA is just an expression tool for vaccine antigens. After mRNA is injected into the body, vaccine antigens are translated into the body. This vaccine antigen induced an immune response, thus bringing protection.
However, the protective power of the vaccine actually comes from whether its own antigen is excellent. It doesn't depend on how the vaccine antigen is expressed.
You use mRNA to inject it into the body, allowing the body to translate the vaccine antigen. Or directly inject vaccine antigen into the body, there is actually no essential difference.
3, why is the inactivated vaccine not good enough?
Because the effective antigen in the inactivated vaccine is the entire S protein of the virus. But this S protein is basically an original structure.
. At present, Pfizer, Moderna vaccine has optimized the structure of the entire S protein and added 2 proline (i.e. S-2P technology). This allows the 3-body structure of S protein to appear in a stable open state. This allows the optimized S protein to induce a better immune response.
One is the optimized S protein, while inactivation is an unoptimized one. The effects are naturally different.
So, the Pfizer vaccine and Moderna vaccine are good at its antigen design (S protein is optimized with S-2P technology), rather than using mRNA input.
4. We can directly inject the optimized S protein as vaccine
In fact, recombinant protein vaccine can be directly injected into the body with the optimized S protein, thereby producing an excellent immune response.
There are already domestic technologies to produce 4-valent recombinant protein vaccines. The vaccine contains S proteins of α, β, Delta, Omicron (optimized).
Because it is a traditional recombinant protein vaccine, its fever after vaccination is much lower than that of mRNA vaccine.
This is the best vaccine possible.
Although the protection effect will not be much different from the existing bivalent mRNA new crown vaccine. Because their antigen structure does not differ much.
It should be pointed out that this 4-price may have special significance for mainland Chinese people. After all, we have not experienced the baptism of the α, β, and Delta virus strains. And it has basically suffered from it overseas. This may have extra meaning for us.
Of course, it may not make much sense.
But think about it, why did Pfizer and Moderna make bivalent vaccines containing Delta+Omicron virus antigen? There are still some concerns.
So, I am actually most optimistic about the 4-valent COVID-19 vaccine that has been optimized as an antigen.
