2 Hydrogen medical concept proposed and evaluation

It is recognized as a selective scavenger of reactive oxygen species (ROS) and especially hydroxyl (•OH), which is related to the occurrence and progression of many pathological states, including cancer.
As early as 1937, hydrogen was first tested as a submersible gas, proving that hydrogen is the best respiratory medium for moderate and deep diving and is extremely safe for the body. did not find any toxic side effects .
It was not until recent decades that the biological effects of hydrogen were discovered one after another. In 1975, American scientists Dole and others reported in the journal Science. After 8 atmospheric pressures of high-pressure hydrogen treatment for skin squamous cell carcinoma mice, they found that the tumor was significantly reduced. This is the first time in human history that hydrogen has a biological effect . However, due to safety considerations, the study did not receive much attention at the time.
In 2007, Oda Naio Oda and others published a paper in "Natural Medicine", reporting that hydrogen has obvious treatment effect on the model of cerebral infarction in rats. Experimental data show that intake of 1%-4% hydrogen can reduce the area of cerebral infarction in rats. They further pointed out that hydrogen has the effect of specifically removing •OH and peroxy nitrite , but does not affect hydrogen peroxide or superoxide, and opens a new chapter in hydrogen medicine. In 2008, Professor Sun Xuejun's team of Professor Sun Xuejun of the Second Military Medical University published the second international article and the first domestic hydrogen medicine research report. Professor Sun Xuejun is the most important advocate and promoter of domestic hydrogen medicine research and is known as "the first person in China's hydrogen molecules."

3 Some important experiments on hydrogen
The earliest report on the biological role of hydrogen was Dole et al. They found that 97.5% of hydrogen was continuously intake at 8 atmospheres. After two weeks, the squamous cell carcinoma of the mouse skin shrunk significantly. They speculated that hydrogen may be a catalyst for free radical reactions. Due to the special experimental conditions and poor repeatability, this study could not be carried out.
Then Gharib et al. found that breathing 7 atmospheres of hydrogen has a significant therapeutic effect on inflammation after parasitic infection in mice. proved for the first time that hydrogen has an anti-inflammatory effect. and proposed that the direct reaction of hydrogen with OH is the basis for its treatment of inflammatory damage, but they also did not carry out any subsequent research work. Although these individual studies have shown some phenomena in the treatment of certain diseases, they have not aroused the interest of other scholars due to unknown mechanisms, poor research, and the need for high-barometric conditions.
2007 Ohsa⁁wa et al. published a paper in Nature Medicine, proving that intake of a small amount of hydrogen (2%) under normal pressure can effectively treat ischemic stroke in rats, and proposed that hydrogen can selectively remove free radical , that is, hydrogen has a selective antioxidant effect. This discovery officially kicked off the study of hydrogen molecular biology.
2008 Nakao and others reported for the first time that continuous intake of 2% hydrogen could prevent peristaltic disorders after small intestine transplantation in rats, significantly enhance the contractility of jejunal smooth muscles, alleviate intestinal mucosal morphological damage, and significantly decrease the MDA level of the end product of lipid peroxidation in the graft. The mRNA of several important inflammatory molecules was determined by quantitative PCR, such as CC group chemokine 2 (CCL2), cytokine interleukin 1β (IL1-beta), cytokine interleukin 6 (IL-6) and tumor necrosis factor α (TNF-alpha), etc. found that hydrogen could significantly inhibit the upregulation of inflammatory factors induced by transplantation.
Then they proved that respiratory hydrogen has a protective effect on heart transplantation injury, lung injury after lung transplantation and chronic transplantation nephropathy. In addition to antioxidant, they proposed that this protection of hydrogen may be related to 's enhancement of the body's anti-inflammatory effects and induction of the antioxidant enzyme HO-1.
Recently, Tan et al. found that as long as hydrogen is dissolved into the organ preservation solution, the gene expression level of apoptotic protein in the heart cells to be transplanted will be significantly reduced, and the oxidative damage will be significantly reduced. The heart is preserved at 4°C in vitro. The time for the heart recovery beating in the control group is about 4 minutes, while the time for the heart recovery beating in the hydrogen-containing protective solution will be reduced to 1 minute, and the effect of heart preservation is significantly improved. The above study shows that hydrogen has the potential application value of organ protection from the storage of isolated organs to the damage after organ transplantation.
